    Flow cytometric analysis of Human PD-1/LAG-3 (Luc) Jurkat Reporter cell staining with PE-Labeled Monoclonal Anti-Human LAG-3 Antibody, Mouse IgG1 (AS659) (Cat. No. FABm061-01) at 1:20 dilution (5 μL of the antibody stock solution corresponds to labeling of 1e6 cells in a final volume of 100 µL), compared with isotype control antibody. PE signal was used to evaluate the binding activity (QC tested).

배경 소개

This PE-conjugated anti-human CD223 (LAG-3) antibody specifically recognizes CD223 (Lymphocyte Activation Gene 3/Protein FDC), a high-performance tool for immunoregulatory research. CD223 (LAG-3) is a ~70 kDa Ig superfamily transmembrane glycoprotein, homologous to CD4, critical for T-cell signaling. Expressed on activated T cells, NK cells and Tregs (higher on CD8⁺ than CD4⁺ T cells), transient in acute activation but sustained under tolerance. As an activation-induced molecule, CD223 binds MHC class II with higher affinity than CD4 (competitive inhibition), associates with TCR-CD3 to downregulate signaling and T-cell expansion (negative T-cell regulation), and promotes dendritic cell activation. CD223 deficiency inhibits tumor growth; synergizes with PD-1 to reverse T-cell exhaustion, enhance tumor rejection and control viral loads. This antibody facilitates CD223 detection, immune mechanism study and tumor immunotherapy research.
