Workflow
Storage
Keep the unopened kit stored at 2-8 °C. Avoid using the kit beyond its expiration date. The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was cal-culated based on the results of the standard curve. The minimum detectable concentration of G-CSF is less than 6.25 pg/mL.
Sample Values
Serum Sample: Thirty healthy donor serum samples were evaluated for the concentrations of human G-CSF in assay.
Cell Culture Supernatant: PBMC (5.0 × 10⁶ cells/mL) were cultured in 1640 supplemented with 10% fetal bovine serum, 100 U/mL penicillin, and 100 ug/mL streptomycin sulfate. Cells were cultured unstimulated or stimulated with 0.1 ug/mL LPS for 2 days or 5 days. Subsequent analysis involved quantitative measurement of human G-CSF concentrations in collected culture supernatants using standardized immunoassays.
Intra-Assay Statistics
Ten replicates of each of three samples containing different G-CSF concentrations were tested in one assay. Acceptable criteria: CV＜10%.
Inter-Assay Statistics
Three samples containing different concentrations of G-CSF were tested in inde-pendent assays. Acceptable criteria: CV<15%.
Recovery
Recombinant G-CSF was spiked into three human serum samples, and then ana-lyzed. The average recovery of G-CSF for serum samples is 101.2%.
