EpCAM분자 배경

EpCAM is also known as CO171A, EGP, EGP40,GA7332, KSA, M4S, MIC18, MK1, TROP1, hEGP2, and is a pan-epithelial differentiation antigen that is expressed on almost all carcinomas as 17-1A(mAb) antigen. Its constitutional function is being elucidated. It is intricately linked with the Cadherin-Catenin pathway and hence the fundamental WNT pathway responsible for intracellular signaling and polarity. The epithelial cell adhesion molecule (Ep-CAM) is known to express in most epithelial malignancies and was reported as a tumor marker or a candidate of molecular targeting therapy. Ep-CAM cross signaling with N-cadherin involves Pi3K, resulting in the abrogation of the cadherin adhesion complexes in epithelial cells was reported. And Epithelial cell adhesion molecule (Ep-CAM) recently received increased attention as a prognostic factor in breast cancer.

FITC-Labeled Human EpCAM / TROP1 Protein, His Tag
I tested the FITC-EpCAM protein for binding to our anti-EpCAM CAR-T cells which were made by lentiviral transduction of human T cells . The protein stained the CAR-T cells but not the control T cells at both 10 ug/ml and 2 ug/ml. It's a good product.

Overall, we were very pleased with the entire process of buying and using the product—shipping was quick and the product was packaged and stored well. The protein was used in the development of a lateral-flow immunoassay (LFA) to detect for EpCAM. Combinations of anti-EpCAM antibodies were first screened using the product, from which we were able to successfully see strong binding on spot tests in duplicate for certain antibody combination pairs. These tests allowed us to confidently move forward with printing and assembling proper LFAs.
