Source

Bat coronavirus BANAL20-236 Spike Trimer Protein, His Tag (SPN-S522d) is expressed from human 293 cells (HEK293). It contains AA Ser 14 - Pro 1205 (Accession # UAY13253.1 (R677A, K978P, V979P)).

Predicted N-terminus: Ser 14

Molecular Characterization

This protein carries a polyhistidine tag at the C-terminus.

The protein has a calculated MW of 137.3 kDa. The protein migrates as >130 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE).

The protein is designed as a trimer.

Endotoxin

Less than 1.0 EU per μg by the LAL method / rFC method.

Purity

>90% as determined by SDS-PAGE.

>90% as determined by SEC-MALS.

Formulation

Lyophilized from 0.22 μm filtered solution in 0.1 M Sodium citrate, pH5.5 with trehalose as protectant.

Background

The spike protein of bat coronavirus BANAL-20-236, a SARS-CoV-2–related sarbecovirus identified in Laos, mediates viral entry by promoting receptor attachment and membrane fusion. BANAL-20-236 shares high genomic similarity with SARS-CoV-2 and contains spike features relevant to cross-species transmission. Specific substitutions, such as P804L in the S2 subunit, have been linked to adaptation to a broad range of Rhinolophus bat ACE2 receptors, making it important for studies on coronavirus evolution and zoonotic spillover risk.
