Storage

For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was calculated based on the results of the standard curve. The minimum detectable concentration of IL-17F is less than 10 pg/mL.
Sample Values
Peripheral blood mononuclear cells from a single donor (PBMCs; seeded at 1 x 10⁶/mL) were cultured in RPMI 1640 supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 ug/mL streptomycin sulfate. Cells were left untreated or treated with 10ug/mL of PHA for 1, 3 and 5days.
Intra-Assay Statistics
Eight replicates of each of 3 samples containing different IL-17F concentrations were tested in one assay. Acceptable criteria: CV＜10%.
Inter-Assay Statistics
Three samples containing different concentrations of IL-17F were tested in independent assays. Acceptable criteria: CV<15%.
Recovery
Recombinant IL-17F was spiked into 3 human serum samples, and then analyzed. The average recovery of IL-17F for serum samples is 97.21%.
